Histological and Immunohistochemical Studies of the Effect of Buspirone Hydrochloride on the Fetal Lung Tissue of Rats

Background and Objective: Buspirone is a pharmacologically categorized anxiolytic that is prescribed to treat anxiety. The sedative medication buspirone is frequently used to treat stress. Buspirone hydrochloride’s effects on fetal lung tissue’s histology and histochemistry have been the focus of the current study. Materials and Methods: Buspirone hydrochloride tablets were obtained from Beecham in Haram, Giza, Egypt. A total of thirty pregnant female rats (12 weeks old, 150-200 g) were separated into 3 groups, each with ten rats. The first group served as the untreated control, in which the pregnant rats were actually given distilled water orally. Group II: Rats received a daily dose of 0.27 mg of buspirone hydrochloride per 100 g of body weight. Group III: The animals were administered 0.41 mg/100 g b.wt./day of buspirone hydrochloride. All doses were administered orally for a period of fifteen days from the 6th-20th day of gestation. The pregnant rats were sacrificed on the day 20 of pregnancy. The uteri were removed before getting the fetuses. The lung tissues of fetuses were taken as samples for histological and histochemical studies. Results: Degenerative alternations included congested bronchial vessels, unequal folds in the mucosa of bronchioles and unequal distributed muscle fibers in the walls of bronchioles were noted in the lung tissues of fetuses of the two treated groups compared to the control group. Also, low content of total protein in the fetal lung tissue, condensed and ridged walls of bronchioles with a decreased staining affinity, alveolar sacs and bronchial vessels. Conclusion: Buspirone hydrochloride exposure by the mother during gestation produced numerous histological and histochemical changes in the lung tissue of fetuses.
